【摘要】 　　目的研究阳和汤对乌拉坦诱导小鼠肺癌的动态影响。方法以乌拉坦(500 mg/kg )1次腹腔注射和雌激素(50 mg/kg)每周1次肌肉注射诱发小鼠肺肿瘤，用阳和汤喂养小鼠18周，每周6次，观察肿瘤生成时间和发生率, 检测其癌变过程中血清和肺组织超氧化物歧化酶(SOD)，过氧化氢酶(CAT)、谷胱甘肽?S?转移酶(GST)的活性及丙二醛(MDA)、白细胞介素?2（IL?2）与肿瘤坏死因子(TNF?α)含量。结果 阳和汤在乌拉坦诱导小鼠肺癌形成过程中对SOD，CAT的活性及IL-2与TNF-α含量有回升作用, 可以推迟肿瘤形成时间，降低肿瘤发生率，并能阻止与癌变相应的GST活性及MDA含量增高。结论阳和汤对乌拉坦诱导的小鼠肺癌有预防作用，主要机制可能与其抗氧化和免疫调节作用有关。

　　Abstract：Objective To study the effect of Yang-he decoction(YHD) on the process of lung carcinogenesis induced by urethane. MethodsThe mouse lung carcinoma was induced by urethane 500 mg/kg injected ip once and diethylstilbestrol 50mg/kg injected im once weekly, the animals were treated with YHD 6 times weekly for 18 weeks to investigate the time and rate of tumor formation，and the activities of SOD ，CAT ，GST and the contents of MDA，IL-2 and TNF-α in serum and in lung homogenate were tested. ResultsIn the process of lung carcinogenesis ,YHD could elevate the activities of SOD and CAT , increase the contents of IL-2 and TNF-α, delay the time of tumor formation , reduce the rate of carcinogenesis , inhibit the increase of GST and MDA due to carcinogenesis. ConclusionYHD can prevent lung carcinogenesis induced by urethane, which might be related to its antioxidantion and immune-regulation .

中医认为,肿瘤的形成是由于机体正气先虚,然后客邪留滞而引起的一系列病变结果。阳和汤是清代名医王洪绪所创，功能温阳补血、散寒逐滞，主治阴疽病症。据文献报道，阳和汤有抗肿瘤作用，可以抑制小鼠S180及HepA实体瘤的生长〔1〕。为探讨肿瘤的发病机制、丰富中药治疗肿瘤的内涵，笔者动态地研究了阳和汤对乌拉坦诱导小鼠肺癌形成的影响。

　　1 材料与仪器

　　1.1 试剂

　　乌拉坦（上海化学试剂厂产品，批号030412）。雌激素（Sigma产品，批号0402134）。SOD、CAT、GST及MDA试剂盒（南京建成生物技术研究所）。TNF-α及IL-2 ELISA 测定试剂盒（华美生物工程公司）。

　　1.2 药物　

　　阳和汤由熟地黄、鹿角胶、白芥子、肉桂、麻黄、炮姜、甘草按20∶6∶4∶2∶1∶1∶2的比例混合, 除鹿角胶外，加入4倍量蒸馏水煎煮1 h，药渣同法再煎煮两次，过滤。 将3次煎煮滤液混合并浓缩至浓度为0.7 g(生药)/ml（浓缩近完成时，将鹿角胶加入烊化），4℃保存备用（一次性购买原药材，分次煎煮，每次煎煮液使用不超过2周）。

　　1.3 动物　

　　昆明小鼠,♀，体重20～23 g（河南医学实验中心,动物合格证号:医动字第410115号）。

　　1.4 仪器

　　EL-800X型酶标仪（BIO-TEK公司）。
