千里光总黄酮的抗炎作用研究(一)

【摘要】 　　目的 探讨千里光总黄酮的抗炎作用。方法通过二甲苯致小鼠耳廓肿胀实验、腹腔注射醋酸致小鼠腹腔毛细血管通透性增高实验以及小鼠棉球肉芽肿实验，观察千里光总黄酮的抗炎作用；制作小鼠气囊滑膜炎模型，测定炎症渗出物中白细胞数（WBC）和前列腺素E2(PGE2)含量。结果千里光总黄酮对二甲苯致小鼠耳廓肿胀、醋酸致小鼠毛细血管通透性的增加以及小鼠棉球肉芽肿的形成均有明显的抑制作用；千里光总黄酮高中低剂量组炎症渗出液中白细胞数和PGE2含量明显低于空白对照组。结论千里光总黄酮对多种炎症模型均有明显的对抗作用，为千里光抗炎作用的主要有效部位之一，且此作用部分是与炎症因子PGE2的产生和释放受抑制有关。

　　Abstract:ObjectiveTo investigate the anti-inflammatory effect of the total flavonoids in Senecio scandens Buch-Ham．MethodsVarious inflammatory models，including the swelling of ear induced by xylene in mice，the permeability increase of blood capillary by acetic acid in mice，and the cotton-pellet granuloma of the mice were used to explore the anti-inflammation of the total flavonoids．Air sac models of synovitis in mice were set up and the amount of white blood cell（WBC）and the content of prostaglandin E2(PGE2) were measured in inflammatory exudan．ResultsThe total flavonoids had significant inhibitory action on auris swell induced by xylene in mice and peration of capillary vessel in mice．The general flavonoids also decreased the proliferation of granuloma caused by implantation cotton pellet in mice significantly．The amount of WBC and content of PGE2 in the experimental groups were obviously lower than those in the control groups．ConclusionThe total flavonoids has significant anti-inflammation，which is relevant to the inhibition of the production and release of the inflammatory factor PGE2．

　　千里光又名千里及、九里明等，系多年生菊科植物千里光Senecio scandens Buch-Ham 之全草，我国南方各省均有分布，主要生长于山坡、林缘、灌木丛中。该药性凉味苦，具有清热解毒、消炎凉血、明目止痛、去腐生肌之功效，临床上主要用于治疗上呼吸道感染、急性扁桃体炎、急性菌痢、急性阑尾炎、急性肠炎、眼结膜炎等〔1〕。植物化学研究表明，黄酮类是该药物的主要成分之一〔2〕，而黄酮类化合物在抗炎、抗变态反应、抗微生物、抗肿瘤等多方面都有一定的药理作用〔3〕。本研究采用多种炎症模型观察了千里光总黄酮(Total flavonoids of Senecio scandens Buch-Ham, TFS)的抗炎作用。现报道如下。

　　1 器材

　　1.1 药物及试剂　

　　千里光购于赣南医学院附属医院中药房。千里光全草切成小段，以体积分数为95%乙醇回流提取3次，提取液合并后减压浓缩除去乙醇，水溶液以50 g・L-1 碳酸钠处理后以氯仿萃取3次，碱水层以1 mol・L-1HCl酸化至pH2后以醋酸乙酯萃取3次，水洗至中性，醋酸乙酯层浓缩成浸膏即得千里光总黄酮（total flavonoids, TF），实验时用灭菌生理盐水(NS)溶解成适当浓度的药液供用。醋酸地塞米松，张家口云峰制药厂（批号20040403）；伊文思蓝，瑞士Fluka公司进口分装（批号20001014），其他试剂均为分析纯。

　　1.2 动物　

　　昆明种小鼠，体重20～22 g，雌雄不限，由江西省实验动物中心提供。

　　1.3 仪器　

　　752紫外光栅光度仪由上海分析仪器总厂生产，高速离心机由上海精密仪器仪表有限公司生产，净化工作台由苏州净化设备厂生产，电子分析天平由上海奇鸿仪器仪表有限公司生产。
